Prokaryotic expression and solubilisation of Arabidopsis ROOT UVB SENSITIVE 1 from inclusion bodies in Escherichia coli

چکیده

The RUS (ROOT UVB SENSITIVE 1) proteins characterized by their unique DUF647 domain are widely distributed in eukaryotes. Their functional roles largely unknown except for the possible involvement of Arabidopsis RUS1 and RUS2 early seedling development. To investigate biochemical proteins, full length truncated were seamlessly fused with GFP cloned into prokaryotic expression vector pQE-100 which allows expressed an N-terminal 6?His tag. Expression RUS1-GFP could not be detected after adding inducer IPTG, while a was at high levels formed inclusion bodies Escherichia coli. dissolved denaturing buffer, then fusion protein supernatant bound to Ni-NTA slurry. eluted non-specific binding washed away. purified as single clear band expected size SDS-PAGE, further confirmed Western blot test. Our results suggest that impossible E. coli can form, effectively solubilized.

Optimizing Primary Recovery and Refolding of Human Interferon-b from Escherichia coli Inclusion Bodies

Background: The refolding of proteins from inclusion bodies is affected by several factors, including solubilization of inclusion bodies by denaturants, removal of the denaturant, and assistance of refolding by small molecule additives. Objectives: The purpose of this study was optimization of recombinant human interferon-b purification in order to achieve higher efficiency, yield, and a produ...

Targeted expression, purification, and cleavage of fusion proteins from inclusion bodies in Escherichia coli.

Today, proteins are typically overexpressed using solubility-enhancing fusion tags that allow for affinity chromatographic purification and subsequent removal by site-specific protease cleavage. In this review, we present an alternative approach to protein production using fusion partners specifically designed to accumulate in insoluble inclusion bodies. The strategy is appropriate for the mass...

FTIR Investigation of Secondary Structure of Reteplase Inclusion Bodies Produced in Escherichia coli in Terms of Urea Concentration

Recent studies suggest that reducing the induction temperature would improve the quality of some recombinant inclusion bodies (IB) by providing a native-like secondary structure and leading to an improvement in protein recovery. This study focused on optimizing the solubilization condition of Reteplase, a recombinant protein with 9 disulfide bonds. The influence of lowering induction temperatur...
